I would say yes, it is possible to connect 2 TMS320DM6446 device via their VLYNQ interfaces.  VLYNQ support bidirectional, peer to peer communication.  I'm not aware of any software implemented that actually does this, but the features of the VLYNQ interface do allow for this capability.

One caution, VLYNQ is intended for device to device communication on a single PCB with very little loading and relatively short trace lengths.  It is not meant for board to board communication, or cabling, etc.

Any recommendations on how the interconnects should be? Can I use an external clock (low frequency) to improve signal integrity? Any other recommendations?

Yes it is possible to communicate two VLYNQ devices on two different boards.I am able to run the two VLYNQ interface on two different boards using cabling.
