We were planning to use OPA604AU op-amp with Ground referenced V+ (40V ) supply.
Can you please suggest me How to handle Offset trim pins of this Op-amp.

If you want to trim and adjust and OPA604's voltage offset, you may use 100kΩ potentiometer and configure the trim circuit as shown in Figure 1. You have approx. +/-50mV trim range as stated.
Or you may leave the OPA604 offset trim pins floating if you don't intent to use the offset adjust function.

As I mentioned in the query, We were planning to connect VCC- to Ground.(Single supply with ground reference).
Is it okay to connect potentiometer between trim points and Ground.

It makes no difference whether you power OPA604 with dual or single supply voltage - the offset trim potentiometer must be connected between the pins 1 and 5 with middle terminal tied to negative rail - see below.
