The biggest difference will be pin 5, on the TCAN1051-Q1 this pin is a no connect, but the SN65HVD1050A-Q1 has the VREF function on pin 5. VREF is meant to drive a stronger common-mode voltage level on the CAN bus to improve stability and thus help with emissions. TCAN1051-Q1 was shown to pass automotive emission standards without the need for this VREF (also called SPLIT) function, so it was not included.

If TI has not sample stock, Can I use TCAN1051-Q1 to replace it directly?
Yes, the TCAN1051-Q1 should replace it just fine, and ultimately be an upgrade from the SN65HVD1050A and the TJA1050.

Yes, even if the customer is actively using the VREF pin, they should be able to directly replace with TCAN1051-Q1.
