Regarding TPD2S017,

  1. What’s the function of the diode in red circle?
  2. How the circuit works if without VCC?

    Please see my answers for your two questions below:

    1.) Those diodes will be the main voltage clamps. Those are the diodes that will breakdown and short the signal to ground during an ESD or surge event. The other diodes are just steering diodes. 

    2.) The Vcc connection is there to lower the line capacitance of the device and also provide additional paths for ESD/surge energy to be dissipated. If there is no Vcc connection, the capacitance and clamping voltage of the device will increase. 

    Also, if you are looking for a 2 channel, 5.5 V device that doesn't have a Vcc pin, I would recommend TPD2E2U06.
