Hi Karl,
HDMI 2.1 the mainly update is that adds support for higher resolutions and higher refresh rates. While TPD5S116 is for voltage level translation, 5V_OUT current limiting and overall ESD protection for the HDMI Controller. From the related features, I don't see difference between HDMI 2.1 and 2.0.
But the HDMI 2.1 was released on November 2017, while TPD5S116 was released on December 2012. So it was not validated for HDMI 2.1. So that still need to check for the HDMI 2.1.
