You have to use the USB Set_Feature command to enable test packet generation.

USB2 compliance test section,

• Reference to section 11.24.2.13 Set Port Feature of the USB2.0 spec

When the feature selector is PORT_TEST, the most significant byte (bits 15..8) of the wIndex field is the selector identifying the specific test mode. Table 11-24 lists the test selector definitions.

USB3 compliance test section,
