Is the USB host able to connect to D1 and USB connector connects to D2? Or it must follow the following connection as below picture?

Hi Randy,

The TUSB216 has bidirectional boost, so yes, you can connect D2 to the connecter and D1 to the host. Please ensure that the DP/DM lanes are still maintaining the correct polarity.
