Can the SUB1 and SUB2 pins of the TPD4S311 connect to USB2.0?
If not, why not?
If so, is SBU1 connected to D+ or SBU2 connected to D+?

Apologies for the delay! Yes, all our protection devices TPD8S300A, TPD6S300A, and TPD4S311A should all be able to support connecting the SBU pin's to D+/D- pins instead. There is no orientation requirement, as the two connector pins (C_SBU1, C_SBU2) are identical and feed to their corresponding system side pins (SBU1, SBU2 respectively). You will just need to connect the corresponding system SBU pin (SBU1 or SBU2) to the correct D+ or D- line of the system side.
