When the VCC power is ON and the pull up power on the SC*/SD* side is OFF,
is it possible for the current on the SCL/SDA side to flow to the SC*/SD* side?
What happens when the VCC power supply is OFF?

Are you asking if the SC*/SD* pins are pulled to 0V what will the current flow be? If so, there is no current flow.
When the device/supply is powered off, then the pass FET is off, and no current can flow through it.

This occurs when SC*/SD* pins become HiZ.
When the device/power supply is turned on, even if either the SC*/SD* side or the SCL/SDA side becomes HiZ, will no current flow to the HiZ side?

If you have a high impedance input, then Ohm's law would show us that the current through the switch would effectively be zero.
