I'm using THVD9491-SEP for one direction only so there is no use for R, A and B pins ( RA will be tied to VCC), Should I leave it floating of tie it to GND?

Thanks you Clemens for answering this question.
You may see some additional leakage current (into Vcc) with A/B floating but the device itself should still work fine.
