Please Let me know the difference between these two transceivers, as per my understanding the first supports half duplex RS232(1T1R mode) and second supports full duplex RS232( 2T2R mode). please confirm this.

As you have noted, the main difference is that the THVD4411 offers 1T1R RS232 mode, where the THVD4421 offers 2T2R RS232 mode.
However, the "half" and "full" duplex configuration terminologies are only related to RS485. The best way to understand this would be compare the THVD4411 datasheet, these three figures: Figure 7-2. (RS-232 Mode), Figure 7-3. (RS-485 Half Duplex Mode) and Figure 7-4. (RS-485 Full Duplex Mode).
Similarly, compare Figure 7-3. (RS-232 2T2R Mode), Figure 7-4. (RS-485 Half Duplex Mode) and Figure 7-5. (RS-485 Full Duplex Mode) in the THVD4421 datasheet:

For L1(Pin number 3) and L2(Pin number 4), should we use UART lines, or can we use GPIOs with pull-up (PU) and pull-down (PD) options?

Feel free to look over our EVM design for the THVD4411: THVD4411 Evaluation Module User's Guide
We implement pullup / pulldown resistors options. You can always leave a footprint and DNI the component if you want flexibility your design.
