TIC12400-Q1: Switch input detection in given fashion

if the switch is connected in above fashion. will we be able to check the switch status (closed and open)?

    Are you asking if you can detect an open or closed switch located between two IN pins, but that is not referenced to GND?

    If so, in theory yes this is possible if one of the IN pins is configured to Source current, and the other IN pin is configured to Sink current.  Only 10 of the devices IN pins are capable of sinking current and you would need to have a larger current sink configuration than you have for the current source pin. 

    This two pin concept is essentially how the Matrix Mode works in the device, so please review that section of the datasheet for more information.  If you only have one switch to connect in this way, then you could create a similar implementation with your register configuration to avoid unused pins in the matrix configuration.
