Hi Xiaoxiang,
Yes, there is only one I2C address from the TUSB216-Q1. You can get a mux to help with the pathing of these I2C lines so that you can program both even though they have the same I2C address. I would recommend looking at the TMUX121: https://www.ti.com/product/TMUX121.
