I don't see any issue with the configuration you have shown.Only point to take care is the that total capacitance on 12 inch cannot be more than 40pF. If this is more then the maximum speed of operation will be proportionally lower.

In relation to this question, what are the concerns in using the configuration below with two ISO1540 devices, where side 2 of both isolation devices is connected to the cable? Since side 2 can drive 400 pf, this configuration should be able to work bidirectionally, correct? Would there be any issues with side 2 of one device driving side 2 of the other device?

I don't see any issues with this configuration. Side2 of ISO1540 is completely I2C compliant. Side 2 of one device can drive Side 2 of another.
