I have a design where I need to route a single USB bus from a Processor through 2 USB switches to a USB HUB and a USB Device Connector. Would this be OK to do using the TS3USB221 (See attached diagram)?

The diagram you show is ok as long as you follow good layout practices to keep the signal integrity of your system. App note for High speed interface layout guidelines.
If you end up needing more bandwidth performance out of your USB switch you can look at the TS3USB3000.
