I'd like to use the TLK1221 in non-sync mode for an application that does not use 8B/10B. What is the minimum transition density requirement for the rx clock recovery?

For applications that do not require framed data the user can disable byte alignment by tying SYNCEN low. This device is compliant with 8b/10b encoding/decoding, hence, if you want to use 64b/66b I would suggest another option such as TLK10031.
