I would like to test the TUSB211 using a signal generator, but I'm having problems activating the hi speed mode with a pseudo random signal. Is it possible to do so?

Please refer to section 8.2.2.1.1 (For Host) or 8.2.2.1.2 (For Device) of the TuSB211 datasheet for specific test procedure when interfacing with test equipment for eye-diagram captures.

I would like to use a signal generator in lieu of the host computer and the TEST_PACKET signal as outlined in the datasheet. When I generate a 480MHz signal, the TUSB211 EVM does not go into Hi speed mode. Is this alternate method possible?

Yes. The sequence required is:
Connect USB test fixture (Terminated at scope)
Configure desired EQ setting on TUSB211.
Power on TUSB211 (With Test fixture connected).
Start sending test packet (This step must be done last).
At this point High-speed should be enabled.
