In default mode, the pin of TCA9535 is input port and I want to know if it is a high-z pin during this period?

Hey John,
Looking at the functional diagram, the device is indeed a high Z input:
Q1 and Q2 are controlled by FETs which are not conducting when the device is configured as an input. The XOR and Flip Flops are typically made using FETs and the inputs into these are the gates of a FET which are high impedance.
