In position1&2, what kind of test equipment and software can be use to analyze the signal quality and reliability?

If you just evaluate the SI, you can use oscilloscope (high speed bandwidth to cover the line rate test) with jitter analysis module.

btw, receiver side has "lock" and "pass" pin indication to help assess the bit error.
