I use TUSB564 in the UFP_D system.

When I use TUSB564 as GPIO mode, is it need to connect HPDIN to DP RX?

DP RX should be connected to the DPXp/n pins. HPDIN should be connected to HPD (Hot Plug Detect) coming from the cable.

When VCC of TUSB564 is 0V, can HPDIN pin apply 3.3V ?

TUSB564 HPDIN should not see 3.3V when Vcc is 0V.
