TUSB1002A: How many levels of IC could be stacked?

You can cascade multiple TUSB1002A together. Do you have customer design info such as block diagram, channel loss, etc so we can figure out what is the best way to support this?

I have a project that requires a similar architecture, and I want to know if the TUSB1002A and TUSB211 can be used in series.
I mean to perform 3 times of direct amplification at the source of the signal?
Is the amplifier inside the IC distorted or saturated?

TUSB1002A is the linear redriver for USB3.1 Gen 2 while TUSB211 is the linear redriver for USB2.0. We have different USB2.0 redriver product, so we need to see which one is the best to fit your need.

Do you have a block diagram and channel loss you can share to show your architecture so we can propose the best place of the redriver? Please note that TUSB1002A and TUSB211 are linear driver, while we can equalize the channel loss, we can't compensate for the DC loss (loss in signal amplitude), so the source has to boost its amplitude to compensate for the DC loss.
