It is posible to work with 8 bit Data In (Single Mode)? If yes, the the first 4 lsb should be '0' ?

Yes, and you can tie any unused bits to ground.

It is matters if I put the 4 LSB or the  4 MSB at '0' ?

You should ground the LSB.
