Due to space constraint i would like to route D+ signal through Pin no 5 (as PIN 5 internally not connected ). It will create any performance or stability problem in the feature?

Pin 5 is not internally connected to the device so it would be okay to route through this pin.
In the layout example in the datasheet they send the signal line D- to the bottom layer because in the example D+ also gets sent to the bottom layer to not connect with pin 6. If D+ and D- get separated it might impose a problem to the system. In your case since D+ stays in the same layer it wouldn't be a problem to route through pin 5.
