Would this device work ok as I2C buffer if we use a 1K pullup on the I"C bus - as this will dominate over the 4K pulldown ?

The device will not work best for I2C (open-drain) as it supports mainly push-pull.
Please see the LSF or TXS family (LSF0204 or TXS0104E) for I2C applications, thanks.

As I want to use the same device for SPI memory buffer and I2C, the TXS104 would be OK?. I realise it has higher thresholds, the SPI wouldnt be running past 1MHZ
Yes, the TXS can be used for SPI. (The TXS has internal pull-up resistors, so you cannot use external pull-down resistors.)
