Unused pin must be held at Vcc or Gnd. Does this mean that we must pull up / down? Can we connect unused pin to Vcc or GND directly?

CMOS inputs can be connected directly to VCC or GND. You need pull-up/-downs only if you want to be able to override the voltage level.
For the SN74LVC245A, it is recommended to ground all unused pins; see [FAQ] What should be done with unused I/O pins of the level translator devices?
