Hi James,
When Vin is provided, internally, each channel detects the short-to-battery by comparing the voltage at the OUT and IN pins before the switch turns on. When Vin is floating, applying a 16V on the output is not likely to damage the part, and I would say the TPS7B770X-Q1 should be able to support this test.

If we use this as high-side switch function, with only 12uA-62uA output current, could this part still perform the open and short diagnostic?

The protection features of the TPS7B770X is still activated when it is used in switched mode.
