I want to use AE11 Pin as Boot Mode and Vout_DE Pin. (MUX)
Boot Mode is used as Input at booting sequence and VOUT_DE changes its function afterwards.
Should I use the Buffer IC applied to the reference schematic?

A buffer is not required. The pin will be an input during reset and after boot. It is recommended to use a pull resistor to set the BOOTMODE to the desired state. Once software has booted and the pin changed to function as VOUT_DE, the pin can function as an output and over-drive the pull resistor.
