Does this mean I-Speed and 370HR share the same layout? Only the stack-up is different?

Because I only find one layout file, and I am not sure it is for I-speed or 370HR.

Yes - same layout.  Its possible the trace widths possibly change slightly with change in PCB stackup/material - but no other changes.

No - the reference design was a study, not a completed/built design.  In the image you shared, there is a Stackups directory that should contain the PCB infomation.
