The word, "internal SPI or I2C connection" means "internal" SPI/I2C connection (interface) between main domain and mcu domain in SOC.
Basically, this interface do not have "physical pin out" on chip.
for example, one SPI interface in MCU domain, and another SPI interface in main domain, this SPI interface connection each other inside chip, but they do not have physical pin out.
Once FFI or MCU isolation enabled, RPMsg could be prohibited, so "internal SPI or I2C connection" is the only way to bridge communication between these two domain.

Can you please specify the exact pins which you are asking about/looking at?
I am also assuming you are talking with respect to custom board?

I have gone through the AM62x TRM and I have not found anything as such which you have highlighted above from MCSPI section.
I have not seen anything as such for MCSPI at the moment based off TRM and datasheet as well. I have also looked at the block diagram but I see no internal connections for MCSPI.

I think it should be not support.
so it should be connect SPI between main/mcu domain by physical pin out.
