What I2C header? Do you mean the EEPROM sequence 0xEE3355AA? This is to identify that a valid EEPROM contents is found.

In the http://processors.wiki.ti.com/index.php/AM335xStarterKitHardwareUsersGuide, u can see I2C header connect to I2C0 in the "functional block diagram" section, is it necessary for the design??

No, this is just a header for connecting external I2C devices or debugging I2C.
