In the reference design of AM335x released by TI the power IC(tps65910) works on EERPOM mode(Boot1 = 1, Boot0 =0) . Does that mean an external I2C EEPROM(i.e. CAT24C256W) must be connected to power IC through I2C interface?

No, the external EEPROM has nothing to do with the PMIC EEPROM mode. TPS65910A EEPROM mode refers to the power sequencing timings that are factory-programmed inside the PMIC EEPROM area.
