The XDS100v2 brought native support for the 64x+ core, including DM643x, as one of its key features. In CCS make sure you choose XDS100v2 and not XDS100. You should then see a choice for EVMDM6437. Go ahead and choose that as the target. The gel file is already in included. Give it a try.
The intent is for the emulator to be independent of the board.
• Emulator •If you're using on-board emulation then you choose "Spectrum Digital DSK-EVM-eZdsp onboard USB emulator".
• If you're using the external XDS100v2 you choose "Texas Instruments XDS100v2 USB Emulator".
• Processor/Board •DM6437 no gel file -- select "TMS320DM6437"
• DM6437 with EVM gel file -- select "EVMDM6437"
If it's your own custom hardware for DM6437 then I would recommend that you copy/paste the gel file from the EVM and make adjustments to customize for your own board, e.g. PLL and DDR settings. Then you can select "TMS320DM6437" go to the Advanced tab, click on the 64x+ core, and then add your new gel file as the one for it to use.

If there is any support on the CD/DVD that came with the EVMDM6437, check if there are emulation drivers there.

Kindly ensure the LEDs, DS501 and DS502 which indicate embedded USB status. DS502 is on when embedded USB emulation is selected and off when the external JTAG emulator is plugged into connector J6. For more details, kindly refer Table 18, Section 3.4 from the tech. ref. doc. below:
