Is the TMS320C6416TBZLZD1 (1GHz) drop-in replaceable for the TMS320C6416TBZLZ7 (720MHz). What would need to be changed to make this replacement. This is being prompted by availabiltiy.

Is this a 1GHz part to be ran at 720MHz (i.e. trying a true drop in) or is this updating a system from 720MHz to 1GHz. You should always check the data manual for differences, I believe the voltage level will be different between the two versions. And yes, even if you're running the 1GHz device at 720MHz you'll still need to operate it at the 1GHz device voltage level specifications. If operating at the higher speed, then the board should also be checked for appropriate clocking and power supply sizing for 1GHz.
Due diligence must be done checking the requirements within the data manual. It should not require significant changes, but some changes will be required.

For 6416T the core voltage is the same for 720 MHz and 1 GHz operation. I looked through the data sheet for several minutes trying to find anything that would need to change, but didn't see anything.

You are correct, 720MHz and 1GHz both have a 1.2V CVDD requirement (the 600MHz had a 1.1V CVDD.) There are differences in typical power numbers between the two units and should be taken into consideration, and if they run at 1GHz the clocking should be looked at to make sure it's correct.
