In section titled: Compiling R5 and ARM64 images
The J721E EVM refers to TDA4VM. Supported modes are: SD/eMMC/OSPI/UART/Hyperflash/USB DFU
No support for PCIe boot.
So the answer is No to both of your questions.

Could you give me an answer to my first question?
If AM6548 supports it, a custom MLO (SBL) can load U-Boot from NVMe M.2 SSD and future SDK releases may support PCIe boot.

The answer i gave is common for both AM654 & J721e. The link i provided above describes
all the supported boot modes from RoM & PCIe is not supported. Hope you can resolve
the question.

The link you provided should indicate that the software (MLO and U-Boot) does not support PCIe boot.
C6748 (hardware and ROM bootloader) supports PCIe Boot as described in TRM.
Does the PCIe Boot support booting from NVMe M.2 SSDs?

I checked with the RoM Expert. ROM configuration for PCIe boot will be endpoint
and it can communicate with Root Complex and will not support boot for PCIe SSD.

U-Boot contains the following source codes, so it should support to access NVMe M.2 SSD.

Booting to Linux kernel is not supported on our SDK. This needs multiple
driver support which are not present.
