I would suggest you connect CC2541EMK to your desktop and run Btool to communicate with CC2640 BLE devices. You can try to refer to processors.wiki.ti.com/.../LPRF_BLE_Simple_Application and www.ti.com/.../swru270c.pdf
If you use CC2541DK (SmartRF05EB+CC2541EMK), you have to download HostTest example to it and connect CC2541DK to your desktop with RS-232 cable. Then, you can start BLE device monitor and it should connect to your CC2541DK.
