Thankyou for the quick response Kai. Just one more clarification around this, from the circuit configuration shared by you it means that we need to operate opamp at +/-2.5V to measure the internal offset voltage? as I am operating it at V+=5V, V-=0  for my design.

No, but it highly simplifies the measurement. You can also do it in a single supplied circuit. Then you can measure the output voltage in reference to the pseudo ground potential.

Also, do you only want to measure the input offset voltage of OPAmp or do you want it to calibrate out?

I want to nullify its internal offset & add the offset externally as per datasheet.

Here i get Vo as 5mV.

I tried the way you have shown with above configuration, i have attached snip below but still i get Vo as 5mV, Can you pls help me around this; with single supply how should i nullify the internal offset?

Hi Jagruti,

you normally use a pseudo ground in such a single supplied differential amplifier, because when both input voltages are identical the output should emit 0V. But it's impossible for the TLV316 to go all the way down to 0V. To omit this unwanted situation a pseudo ground is usually introduced at the ground terminal of R2.
