Yes, the INA241 should be operable at -3V but the NC should be left unconnected to avoid creating ground loops or short circuits.

The NC pin is not connected internally to the die, so in general, connecting it to ground would help improve heat spreading. Since you are connecting GND to -3V it is advisable for your NC to be left floating.
There are no limitations to the operating condition, just ensure that all your components can function at -3V.
