Is it possible a single +5V supply on the ISO124 by connection VS1- to GND1 and VS2- to GND2 and their respective supplies to +5V, when the input is limited to 0-5V?

Yes but you will have very limited signal swing. This is not a rail-to-rail device. Typical input and output swing performance is 2.5V to each rail so you might get reasonable performance with signals from 2.5V to 9.5V. Tested and assured performance is 5V to the rails (a conservative rating), or 5V to 7V.
