We now design a measurement scheme (as shown below), measure the THDN of the audio amplifier output signal with TLV320ADC3140. Please help your suggestions. Thank you!

I don't see anything wrong with your plan to do this. I'm assuming the THDN you are measuring is going to be higher than that of the ADC3140. Just make sure to respect the input range, the integrated PGA gives you a lot of flexibility though. Let us know if you have any questions or issues.

The TLV320ADC3140 actually has an integrated PGA that can apply up to 42dB of analog gain in addition to a digital volume control that can apply up to 27dB of digital gain. To take full advantage of this range I would recommend AC coupling the inputs. I would also note that the integrated PGA can only boost, not cut so if that is a feature you need then it may be best to attenuate the signal beforehand or use an external PGA.
I'm personally not familiar with the PGA portfolio so you may want to post another question for PGA recommendations so that the right people will see it.
