ADS1299-6: Difference Vref at bipolar supply to unipolar supply

There is a nice explanation from you about the reference voltage, resolution and  convert these to code.

I used a +-2.5 powersupply and connect the Vref like in the picture. It's working fine. The Vref measured with a voltmeter is 4.5V from Pin24 to Pin25.

Only my value  is 4 times bigger than in your example. Do I have to pay attention to this type of reference?

    With the information provided I'm not really sure what would be causing this problem.
    What does your current equation look like?
    Remember that one of the 2's from this equation: (2 x VREF)/ Gain / (2^24 - 1) get's canceled out.
    Ensure that the registers for gain and external reference are properly configured.
