1. When using 5V tolerant IO, there is no need to input a separate 5V power supply, right?
2. Since MSPM0 has two tolerant IOs, does it support I2C and UART of 5V?

They are both open drain IOs, so if you want to use them at 5V you do need a 5V rail to pull them up to, but you are correct that there's no need to supply 5V to the device itself.

Yes, that is correct. PA0 and PA1, which are the 5V ODIOs, have UART and I2C peripherals available on them. So you can use these with an appropriately sized pullup and support I2C or UART.
One thing to be aware of, is that if you are in a 20-pin or smaller package, then NRST is bonded internally to PA1. So in those packages the PA1/NRST pin is not 5V tolerant. PA0 can still operate up to 5V though.
