2. What do the marked blue and marked red mean?

Hi Enhui,

1) It's possible that noise can couple across to the analog input but not likely to the clock due to the distance.

2) Red is Analog input side Blue is Digital output

Whether the clock will effect the signal in the analog area ? If you have any suggestions of the PCB layout? Thanks.

Hi Enhui,

If you are concerned about the clock coupling in then the clock should be routed on a different layer than the analog inputs
