Hi Kai,
Yes, TPS53625 will work with 5V Vin.. But be carefull to select right Inductor and Vin/Vout cap's at powerstage input/output.

Yes, you need a 3.3V bias rail for VDD input pin that can be generated from the 5V power input.

Please ensure you select the right Inductor and Vin/Vout capacitors for the powerstage input/output.
