Can I use TP10(Data), TP9(CLK), TP7(STROBE), TP6(RDBK) to program TRF3765 PLL not using FT245RL?

Hi BH,

Yes, this is possible.

First of all, do not connect the USB to a PC. Without power supply, FT245RL will keep its output Hi-Z. You are safe to connect the SPI test points to an external MCU.
