By looking at your schematic it looks like you are using DP input and not DP++ input into the SN75DP159.
The SN75DP159 is a DP++ to HDMI 2.0 retimer and not a DP to HDMI active protocol converter.
DP++ uses HDMI protocol encoding or TMDS and therefore the SN75DP159 is an passive protocol converter.

You will need an active protocol converter to take the DP signal and convert it into TMDS signaling into the input of DP159.

I checked with our graphics module provider, the interface is DP++.
We are not getting any transactions on SDA_SRC, SCL_SRC. They are always high.

Also, the I2C over AUX functionality is only available in the RGZ package. For the RSB package, the AUX lanes should not be connected to the DP159RSB.
Is the R1699 resistor installed or is the R1695? If you would like I2C enabled you will need only the pull up resistor.
Does the GPU in this application support clock stretching?
Please refer to Figure 31. Implementation for Motherboard 2 of the datasheet for an illustration of an example schematic.
