I want to design IR Sensor using OPT3101. Is distance value will be available on I2C
or is there any need to set registers.

Distance is read from I2C and yes OPT3101 needs to be configured and calibrated before data can be read. Please see here for our collateral on OPT3101

Also want to confirm OPT3101 is a time of flight distance sensor. It can use IR light to do this, but it is not an IR light sensor in the sense that it is not an IR light intensity sensor.

Does OPT3101 needs two supply 1.8V and 3.3V for operating.

OPT3101 has internal LDO mode that allows single supply option. Yes, OPT3101 can work with any MCU with I2C.
